Benadryl (diphenhydramine) is an antihistamine that acts to minimize the chemical nature of histamine in the body. Histamine causes the body to react in several ways, including runny nose, watery eyes, itching, and sneezing. Benadryl is used to treat these symptoms, including skin rash, hives, and other allergy and cold symptoms (Wong, 2015).

The two largest antihistamine classes are the H1 and H2 antihistamines; Benadryl is in the former category. The H1 antihistamines work by bonding the receptors of H1 histamines found in the endothelium, smooth cells, and mast cells in the body. They are also found in the brain’s tuberomammillary nucleus. The H1 receptor target antihistamines are used to treat allergic reactions that occur in the nose, such as sneezing, runny nose, and itching. Additionally, these antihistamine classes are used to treat vertigo, motion sickness, and insomnia caused by inner ear problems (Hu et al., 2015).

The histamine receptors exhibit constitutive activity, so antihistamines can function as inverse agonists or receptor antagonists at histamine receptors. Different H1 antagonist antihistamines carry the brand name of Benadryl in different regions; for example, acrivastine is the active Benadryl Allergy Relief component, while Benadryl One a Day Relief active component is cetirizine in the UK. In the US and Canada, diphenhydramine is the active component of Benadryl.

According to Goldberg and Ernst (2018), a person should stop using Benadryl when they experience tightness in the jaw or neck and uncontrollable tongue movement; feeling like passing out or confusion; no or little urination; difficult or painful urination; and fluttering or pounding heartbeats. The side effects of Benadryl include daytime drowsiness or a feeling of a hangover after using it at night; blurred vision or dry eyes; upset stomach or constipation; dry throat, nose, or mouth; loss of coordination; drowsiness or dizziness. The symptoms should subside after 7 days.
